His parents are dead. It's pretty strongly intimated that they're dead, anyway. Or they're people he has cut off every form of contact with - unlikely, because he's John. There's a scene when Sherlock is first deducing things about John and one of his deductions is that John doesn't have any close relatives outside of his sister, Harry. Ergo. No parents. Plus, they didn't turn up at his wedding, so that's another tip off.
As for what they do, I imagine they were middle-class working people. His father would have probably worked in a factory or construction in a management position, his mother would have had an office job as administrative staff at a hospital. His mother was patriotic and firm, but fair. John takes after her. His father was a stolid man who liked to take John to movies or on fishing/hunting trips, which is where John developed his sharp-shooter skills.
Favorite childhood memory would be going to see James Bond in theaters for the first time with his father and them playing spies after it. One of the few times that his dad was a little less serious. Least favorite memory would be when his sister first came out and she was getting bullied for it. John tried to step in and got pounded into the ground while Harry had to watch and the bullies were yelling homophobic insults at the both of them. That was probably the point that he decided he was going to become better at fighting than every bloke bigger than him to ensure that didn't happen again.
